The Angiotensin II/transforming growth factor-β1 (AngII/TGF-β1) signal axis is an important regulatory pathway for atrial fibrosis, which can contribute to atrial fibrillation (AF). Fused in sarcoma (FUS) was recently found to regulate cardiac diseases. This study aimed to investigate whether FUS could regulate AngII induced fibrosis and uncover the possible mechanisms. The expression of FUS in AF patients and AngII-induced cardiac fibroblasts was measured by RT-qPCR and western blot assays. Fus was silenced in cells using short hairpin RNA (shRNA), then cell proliferation, migration, collagen synthesis and TGF-β1/Smad signaling were detected by CCK-8, wound healing and western blot assays, respectively. The possible target for Fus was predicted by searching Starbase database and verified by RNA-binding protein immunoprecipitation (RIP) and RNA pull down.

Cells were overexpressed with Pax3 in the presence of Fus silence and AngII stimulation, then the above cellular processes were further evaluated. Results showed that FUS was upregulated in AF patients and AngII-induced cardiac fibroblasts. Fus knockdown inhibited AngII-enhanced cell proliferation, migration, collagen synthesis and TGF-β1/Smad signaling activation. Furthermore, Fus functions as an RNA-binding protein to bind to Pax3 mRNA and positively regulate its expression. Further studies demonstrated that Pax3 overexpression canceled the above effects of Fus knockdown on cell proliferation, migration, collagen synthesis, and TGF-β1/Smad signaling activation in AngII-induced cells. In conclusion, Fus could target Pax3 to increase the pro-fibrotic effect of AngII in cardiac fibroblasts via activating TGF-β1/Smad signaling. Knockdown of Fus/Pax3 axis may provide a potential therapy for relieving AF.

Description: Fibroblasts are mesenchymal cells derived from the embryonic mesoderm. They have been extensively used for a wide range of cellular and molecular studies. This is mainly because they are one of easiest types of cells to grow in culture, and their durability makes them amenable to a wide variety of manipulations ranging from studies employing gene transfection to microinjection. There is good evidence that fibroblasts in different parts of the body are intrinsically different. Fibroblasts within tissues are exposed to a dynamic mechnical environment, which influences the structure integrity of both healthy and healing soft tissue. Fibroblasts secrete a nonrigid extracellular matrix that is rich in type I and/or type III collagen They are responsible for much of the synthesis of extracellular matrix in connective tissues and play major roles in wound healing. Many diseases are associated with fibroblasts, either because fibroblasts are implicated in their etiology or because of the fibrosis that accompanies damage to other cell types in tissues. Description: Human Kidney Fibroblasts are isolated from the human adult healthy renal tissue. HKF-a are cryopreserved after passage one culture and delivered frozen. HKF-a are guaranteed to further expand for 15 population doublings under the conditions provided by in the technical sheet. As an excretory organ in the urinary system, the kidney greatly influences an individual’s well being by preserving homeostasis through regulating electrolytes, pH, and blood pressure among other control mechanisms. Comprising the majority of interstitial cells in the kidney, renal fibroblasts are responsible for the synthesis of collagen I of the extracellular matrix. Also, renal cortical fibroblasts synthesize erythropoietin, an important glycoprotein hormone that controls the production of red blood cells and renal medullary fibroblasts produce prostaglandins, a class of autocrine or paracrine hormones, which help maintain water and electrolyte homeostasis. Renal interstitial fibrosis, a disorder that may escalate to chronic kidney disease, is often correlated with excessive deposition of extracellular matrix by renal fibroblasts, the regulation of which process is under intensive research.

Human Cardiac Fibroblasts

ABC-TC3533 1 vial Ask for price
Description: The cardiac fibroblasts (CF) are the principal cell type of the heart. They provide structural support for cardiac myocytes and are responsible for extracellular matrix synthesis in the heart during growth and pathophysiological conditions. CF are an important cellular component of myocardial responses to injury and the source of paracrine growth factors. Their proliferation and synthesis of matrix is essential for scar formation at sites of myocardial infarction, cardiac fibrosis, and is often complicated by cardiac hypertrophy. CF also respond to changing environments in pathophysiological conditions by modulating integrin expression. CF culture has been widely used as a model to study the cardiac matrix remodeled by physiological (exercise) and pathological (hypertension) stressors.HCF from Gentaur Research Laboratories are isolated from the human heart. Description: The most abundant cell type in lung interstitium is fibroblasts. They resemble ordinary fibroblasts but have some distinguishing features, for example, they have long branching processes and gap junctions.

Human Gastric Fibroblasts

ABC-TC5516 1 vial Ask for price
Description: Gastric fibroblasts (GF), which are located in the stomach, are mesenchymal cells derived from the embryonic mesoderm. Fibroblasts secrete a non-rigid extracellular matrix that is both rich in type I and/or type III collagen. They are responsible for much of the synthesis of extracellular matrix in connective tissues and play a central role in wound healing. Studies in vitro and in vivo show that GF can regulate epithelial regeneration by influencing epithelial cell proliferation and differentiation. In addition to their role in wound healing, human GF (HGF) have been shown to be key contributors in the development and progression of gastric carcinoma. HGF are a useful model for examining the underlying mechanisms that cause gastric carcinoma and may provide novel insights for developing new cancer drug therapies.

Human Gingival Fibroblasts

ABC-TC3627 1 vial Ask for price
Description: Fibroblasts are mesenchymal cells with many vital functions during development and in adult organisms. They are responsible for much of the synthesis of extracellular matrix in connective tissues and play major roles in wound healing. Many diseases are associated with fibroblasts, either because fibroblasts are implicated in their etiology or because of the fibrosis that accompanies damage to other cell types in tissues. Fibroblasts are among the most accessible normal mammalian cell types and still the most amenable to culture in vitro. Gingival fibroblasts are the major constituents of gingival tissue and play a key role in their health maintenance. Human gingival fibroblasts (HGnF) express a wide surface molecular panel including mainly CD9, CD26, CD55, CD59, CD63, CD71, CD86 CD95, CD99 and CD117. They were also demonstrated to express mRNAs for protease-activated receptor-1 (PAR-1) and PAR-3.HGnF from Gentaur Research Laboratories are isolated from human gingiva. HGnF are cryopreserved at passage one and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HGnF are characterized by their spindle morphology and immunofluorescent method with antibody to fibronectin. HGnF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HGnF are guaranteed to further expand for 15 population doublings at the condition provided by Gentaur Research Laboratories.

Human Prostate Fibroblasts

ABC-TC3765 1 vial Ask for price
Description: Fibroblasts are mesenchymal cells derived from the embryonic mesoderm. They have been extensively used for a wide range of cellular and molecular studies. This is mainly because they are one of the easiest types of cells to grow in culture, and their durability makes them amenable to a wide variety of manipulations ranging from studies employing gene transfection to microinjection. There is good evidence that fibroblasts in different parts of the body are intrinsically different. Fibroblasts within tissues are exposed to a dynamic mechnical environment, which influences the structure integrity of both healthy and healing soft tissue. Fibroblasts secrete a nonrigid extracellular matrix that is rich in type I and/or type III collagen. Cultured prostate fibroblasts synthesize FGF-like growth factor that stimulate their growth and is believed to be important in the development of benign prostatic hyperplasia.HPrF from Gentaur Research Laboratories are isolated from human prostate tissue. HPrF are cryopreserved at primary culture and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. Human Pulmonary Fibroblasts

ABC-TC3776 1 vial Ask for price
Description: The most abundant cell type in lung interstitium is fibroblasts. They resemble ordinary fibroblasts but have some distinguishing features, for example, they have long branching processes and gap junctions. Their principle function is production of type III collagen, elastin, and proteoglycans of the extracellular matrix of the alveolar septa. Pulmonary fibroblasts (PF) play an important role in the repair and remodeling processes following injury. The controlled accumulation of fibroblasts to sites of inflammation is crucial to effective tissue repair after injury. Either inadequate or excessive accumulation of fibroblasts could result in abnormal tissue function. For example, the excess proliferation of fibroblasts contributes to the adventitial thickening observed during the development of hypoxia-induced pulmonary hypertension. HPF from Gentaur Research Laboratories are isolated from human lung tissue. HPF are cryopreserved at primary culture and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. There is good evidence that fibroblasts in different parts of the body are intrinsically different. Fibroblasts within tissues are exposed to a dynamic mechanical environment, which influences the structure integrity of both healthy and healing soft tissue. Fibroblasts secrete a nonrigid extracellular matrix that is rich in type I and/or type III collagen. L’acide salvianolique B inhibe la différenciation des myofibroblastes et l’accumulation de matrice extracellulaire dans les fibroblastes polypes nasaux via la voie de signalisation TGF-sign1

In the process of nasal tissue remodeling, nasal fibroblasts serve an important role via myofibroblast differentiation and the production of extracellular matrix (ECM). Nasal fibroblast abnormalities can lead to conditions such as chronic rhinosinusitis. Salvianolic acid B (Sal B), a water-soluble active pharmaceutical compound extract from the root of the traditional Chinese medicine Salvia miltiorrhiza, displays antioxidative, antiproliferative and antifibrosis properties. The present study aimed to investigate the mechanism underlying the effects of Sal B on nasal polyp fibroblast (NPF) myofibroblast differentiation and ECM accumulation. Primary NPFs were obtained from nasal polyps of patients with chronic sinusitis. The proliferative and cytotoxic effects of Sal B on NPFs were evaluated by performing the Cell Counting Kit-8 assay.

The Transwell assay was conducted to assess cell migration. α-smooth muscle actin (α-SMA), TGF-β1 receptor (TβR)-I, TβR-II, Smad2/3 mRNA and protein expression levels and (p)-Smad2/3 phosphorylation levels were measured via reverse transcription-quantitative PCR and western blotting, respectively. Type III collagen and fibronectin levels were analyzed by ELISA. The results indicated that Sal B significantly downregulated TGF-β1-induced α-SMA, fibronectin and collagen III expression levels in NPFs. Similarly, Sal B significantly decreased TGF-β1-induced TβR-I, TβR-II, p-Smad2/3, MMP-2 and MMP-9 mRNA and protein expression levels in NPFs. Furthermore, Sal B significantly decreased TGF-β1-induced NPF migration. Therefore, the present study indicated that Sal B inhibited myofibroblast differentiation and ECM accumulation in nasal fibroblasts, suggesting that Sal B may inhibit nasal polyp formation via certain mechanisms.

E2F2 stimule l’expression de CCR4 et active les cellules de type fibroblaste synovial dans la polyarthrite rhumatoïde

Aim of the study: E2F transcription factor 2 (E2F2) has increased expression in synovial tissues of rheumatoid arthritis (RA) and stimulates interleukin (IL)-1 α and IL-β production in cultured RA synovial fibroblast-like cells (RASF), which supports the importance of E2F2 in RA pathogenesis. This study investigated the effect and mechanism of E2F2 in RA.

Material and methods: Cultured RASF were transfected with anti-E2F2 siRNA, and the expression profile was analyzed with an inflammatory response and autoimmunity PCR array loaded with 84-relative genes to explore the pathogenic pathway of E2F2. Apoptosis, migration and tube-like structure formation in the RASF with transfection of anti-E2F2 siRNA or E2F2-expressing plasmids were examined using flow cytometry, transwell assays and Matrigel assays, respectively.

Results: Significantly decreased expression of chemokine receptor 4 (CCR4) was detected in RASF with inhibited E2F2 expression, and the CCR4 expression was increased in RASF with transfection of E2F2-expressing plasmids. Silencing E2F2 expression stimulated apoptosis, but retarded migration and tube-like structure formation in RASF. The opposite observation was obtained in RASF with E2F2 overexpression.

Conclusions: High E2F2 expression decreases apoptosis and increases migration and tube-like structure ability in RASF and might perform this role by up-regulating CCR4 expression, which ultimately contributes to the disease progression of RA synovial tissues.

Taux de sécrétion et composition protéique des vésicules extracellulaires libérées par les fibroblastes associés au cancer après irradiation

Reciprocal communication between the malignant and non-malignant cellular elements in tumors is essential for cancer sustainability and plays an important role in the response of cancers to treatments. Some of this cellular crosstalk takes place via secretion of vesicles that are actively released into the extracellular space by most cell types in tumors. Recent studies have demonstrated radiation-induced changes in the secretion rate and composition of extracellular vesicles (EVs), with impact on radiation-related cellular communication.

However, little is known about the effects of different radiation regimens on the release of EVs by cells of the tumor microenvironment. In this study, we provide a comprehensive molecular characterization of EVs released by cultured primary lung tumor fibroblasts. We explore the quantitative and morphological changes triggered by ionizing radiation (IR), delivered as a single dose of 18 Gy or three consecutive daily medium-doses of 6 Gy. Cancer-associated fibroblasts (CAFs) secrete EVs with sizes ranging from 80 to 200 nm, expressing some of the classical exosome markers. Exposing CAFs to a single-high radiation dose (1 × 18 Gy) or fractionated medium-dose did not alter the release of CAF-EVs.

The protein composition of CAF-EVs was analyzed by LC-MS/MS proteomics and revealed that CAF-EVs are enriched with heat shock proteins, integrins, tetraspanins, proteinases, collagens, growth factors and an array of molecules involved in the regulation of cell migration and the immune system. Quantitative proteomic analyses revealed minor changes in the protein composition of CAF-EVs after radiation exposure. Taken together, this study presents original data on lung tumor CAF-EV composition and reveals that release and protein cargo of CAF-EVs are largely unaltered after exposing CAFs to IR.

HCF-a are cryopreserved at secondary culture and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HCF-a are characterized by immunofluorescent method with antibody to fibronectin. Human Lymphatic Fibroblasts

ABC-TC3688 1 vial Ask for price
Fibroblasts secrete a nonrigid extracellular matrix that is rich in type I and/or type III collagen. Besides, dermal fibroblasts also secrete large quantities of hyaluronan in response to inflammatory stimuli. During wound healing, dermal fibroblasts switch from a migratory, repopulating phenotype to a contractile, matrix-reassembling phenotype.HLF from Gentaur Research Laboratories are isolated from human lymph nodes. HLF are cryopreserved at primary culture and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HLF are characterized by their spindle morphology and immunofluorescent method with antibody to fibronectin. HLF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HLF are guaranteed to further expand for 15 population doublings at the condition provided by Gentaur Research Laboratories.

HPF are characterized by immunofluorescent method with antibody to fibronectin. HPF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HPF are guaranteed to further expand for 15 population doublings at the condition provided by Gentaur Research Laboratories.

Human Esophageal Fibroblasts

ABC-TC3613 1 vial Ask for price
Description: Esophageal fibroblasts which line the lamina propria of the upper and lower esophagus are mesenchymal cells that originated from the embryonic mesoderm. They have been shown to play an important role in angiogenesis through TGF-β and VEGF signaling pathways in the highly angiogenic esophageal squamous cell carcinoma. Predominantly, fibroblasts secrete a nonrigid extracellular matrix that is both rich in type I and/or type III collagen. They are responsible for much of the synthesis of extracellular matrix in the connective tissues and play major roles in wound healing. Because they are one of the simplest types of cells to grow in culture fibroblasts have been extensively used for a wide range of cellular and molecular studies. In addition, fibroblasts’ durability makes them amenable to a wide variety of manipulations ranging from gene transfection to microinjection. The HEF cells from Gentaur Research Laboratories are isolated from human esophageal tissue. HEF are cryopreserved at passage one and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HEF are characterized by immunofluorescence with antibody specific to fibronectin. HEF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HEF are guaranteed to further expand for 15 population doublings at the conditions provided by Gentaur Research Laboratories.

Fibroblasts secrete a non-rigid extracellular matrix that is rich in type I and/or type III collagen. They are responsible for much of the synthesis of extracellular matrix in connective tissues and play major roles in wound healing. Many diseases are associated with fibroblasts, either because fibroblasts are implicated in their etiology or because of the fibrosis that accompanies damage to other cell types in tissues. For example, the development of bowl stenosis in Crohn’s disease patients is caused by extreme fibroblast proliferation and extracellular matrix expansion.HIF from Gentaur Research Laboratories are isolated from human intestinal tissue. HIF are cryopreserved at passage one and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HIF are characterized by their spindle morphology and immunofluorescent method with antibody to fibronectin. HIF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HIF are guaranteed to further expand for 15 population doublings at the condition provided by Gentaur Research Laboratories.

They are responsible for much of the synthesis of extracellular matrix in connective tissues and play major roles in wound healing. Human gallbladder fibroblasts (HGBF) are responsible for eicosanoid synthesis, specifically PGI2. Chronic stimulation of HGBF results in up-regulated synthesis and release of PGI2 which aids in formation of early acute cholecystitis. HGBF are very versatile cells and could be used in studies of many conditions, such as the early stages of acute cholecystitis.HGBF from Gentaur Research Laboratories are isolated from human gallbladder tissue. HGBF are cryopreserved at passage one and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume.
